CHAPTER 1 INTRODUCTION Several unicellular microorganisms use smallsignaling molecules to find out their local concentration. The processes involved in the production and recognitionof these signals are generally known as quorum sensing (QS). Unicellular microorganisms to manage their activities use this kind of cell-to-cell communication, which allows them to work as multi-cellular systems. Newly, several groups have confirmed artificial intraspeciesand inter-species communication through synthetic circuits, which incorporate components of bacterial QSsystems. Engineered QS-based circuits have a broad range of applications such as production of biochemicals, tissueengineering, also mixed-species fermentations. They are also extremely useful in designing microbial biosensors toidentify bacterial species present in the environment andinside living organisms. In this articlethe different ways inwhich researchers have designed QS-based circuits andtheir applications in biotechnology are explained. Detailed explanation ofcurrent progress in building QS-based microbial biosensors and QS-based biocontrol are given. Lastly, discussion of QS inhibition as a viable strategy for the decline of biofouling is given. Also different applications of QS in biotechnology are given. Engineered Quorum Sensing systems The promisingfield of synthetic biology seeks to generatenovel biological systems by applying the fundamental engineering principles of standardization and hierarchical abstraction to GE engineering (Purnick and Weiss 2009). The finding that many pathogens rely on cell-to-cell communication mechanisms, known as quorum sensing, to synchronize microbial activities essential for infection and survival in the host suggests a promising disease control strategy, i.e. quenching microbial quorum sensing or in short, quorum quenching. Work over the past few years has demonstrated that quorum-quenching mechanisms are widely conserved in many prokaryotic and eukaryotic organisms. These naturally occurring quorum-quenching mechanisms appear to play important roles in microbe-microbe and pathogen-host interactions and have been used, or served as lead compounds, in developing and formulating a new generation of antimicrobials. An advance study of bacterial quorum sensing process can facilitate development of novel technologies intended at interfering with bacterial communication and virulence. The term quorum sensing explains the capability of a microorganism to recognize and response to diffusible signal molecules. As mentioned in above applications of QS there are many areas that are fully touched by QS method. Therefore, significant research efforts are needed to understanding Quorum Sensing and the growth of strategies to disrupt and influence Quorum Sensing. Our understanding of quorum-sensing mechanisms currently restricts applications for quorum sensing. Though there has been progress made in the use of quorum sensing, more understanding of quorum functionality is necessary before the control of this tool can be completely raised. However, the full-scale management of the bacterial quorum circuit in a biotechnological application yet to be an unconvinced goal. More studyand deep research is needed to uncover andthe details of QS in a diversity of microbial species, with Gram- positive bacterias and fungis. The task of QS in microbial populations, with Quorum Sensing crosstalk and signal specificity, is another significant area of research and study that will influence strategies to prevent biofilm formation and for biocontrol. Quorum sensing seems to be a distinctive example of how the exploitation of bacteria cell-to-cell communication in biotechnology can be used to significantly drive the growth and development of medicine, diagnosis tics, therapies and gene control. For sure, it will influence every part of biology, with novel research and technologies in science world.

An Analysis of Freytag’s Five Steps in Shakespeare’s “The Tempest” Essay

In this essay I will analyze William Shakespeare’s â€Å"The Tempest† using Gustav Freytag’s five phases which are: exposition, point of conflict, rising action, climax, and denouement.   I will first begin with the exposition phase.   The exposition phase includes a description of the chief protagonist and antagonist as well as a description of their conflict and setting.   I will also pinpoint the inciting moment, which is an incident that must occur for the story to happen. (www.reference.com)   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   The chief protagonist of the play is Prospero, the rightful duke of Milan.   The main antagonist is his brother, Antonio, the usurping duke of Milan.   Antonio overthrew Prospero and sent him and his young daughter Miranda out to sea.   They land on an island, which will be the central setting of the play, where Prospero learns magic and frees a spirit named Ariel who is indebted in servitude to him.   Prospero uses his magic to stir up a storm and moor a ship that his brother Antonio is on.   Ariel makes sure that the occupants of the boat arrive on the island unscathed.   The inciting incident is the arrival of Antonio and the rest of his party on the island.   The main conflict is between Prospero, who wants his dukedom reinstated, and Antonio, who wants the dukedom for himself.   Next, I will describe the rising action in the play.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   The rising action includes related secondary conflicts. (www.reference.com)The first of these secondary conflicts occurs between Antonio and Sebastian, the king of Naples brother, and Gonzalo, a counselor, and Alonso, the king of Naples.   These gentlemen had been traveling together on the boat, but when they come to the island Antonio and Sebastian plan to murder Alonso and Gonzalo so that they can seize the power of the crown for themselves.   Alonso believes his son, who had been traveling with them as well to be dead and Antonio and Sebastian see this as a chance to eliminate Alonso and usurp the crown.    Alonso’s son Ferdinand really isn’t dead and is busy falling in love with Prospero’s daughter while this is happening.   The last secondary conflict begins with Caliban, Prospero’s unwilling slave, and Stephano and Trinuculo, a jester and a drunken butler who were traveling with the party on the boat, meeting each other on the island and deciding to kill Prospero.   Ariel hears this and informs Prospero thus avoiding catastrophe.   Now, I will discuss the falling action, climax, and denouement.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   The climax takes place in â€Å"The Tempest† when Prospero charms the party from the boat and Caliban, Stephano, and Trinuculo into immobility.   Prospero, who is joyous at the thought of his daughter’s upcoming marriage to Ferdinand, has a change of heart and releases everyone from the spell.   They are all repentant and the falling action occurs at this point with a monologue from Prospero chastising the king of Naples and Antonio for the part the played in the usurping of his dukedom. He also chastises his slave and the butler and jester for trying to kill him.   The denouement encompasses the happy marriage of Miranda and Ferdinand and the reinstatement of Prospero’s dukedom.   The play ends happily with the protagonist better of at the end than at the outset because it is a comedy rather than a tragedy.   Thus, I have analyzed â€Å"The Tempest† and have discerned Freytag’s five phases within the play. An Analysis of J.D. Salinger’s The Catcher in the Rye

In J.D. Salinger’s The Catcher in the Rye, Holden Caulfield is portrayed as a young, troubled individual. He tells us his story from the mental institution where he is currently residing. Holden is a 16 year old going through many different adolescent changes. He is expelled from his prep school for flunking too many subjects. He drinks, smokes, sees a prostitute, is punched by her pimp, goes on dates, spends a great deal of time in the park, and really does not do a great deal else. Holden is a very hard person to figure out and analyze. Throughout the whole book, Holden constantly changes his mind about things and has various conflicting thoughts. One of the biggest reasons is the fearing of growing up and getting older because getting older means you are closer to death (Hennessy, 2010). Holden decides to â€Å"numb himself† to his feelings about life to the point where it seems like he is no longer truly living but is just going through the motions day by day. Going through adolescence can be a difficult time. At some times it may feel like everyone is against you. This is similar to what Holden experienced in Catcher in the Rye. He continuously tried to alienate himself from society; he suffers from a self-imposed delusion that he is different. Throughout the story, Holden realizes that growing up indicates that he should assume responsibility in a world where he thinks only superficial and insignificant issues occur (Abdulatif, 2010). Surfus decided to incorporate this nonprofit formally after experiencing sexual orientation discrimination at a local business in Grand Rapids, MI.
